產(chǎn)品簡介:
鐵死亡(Ferroptosis)是一種鐵依賴形式的非凋亡性細(xì)胞死亡。Ferrostatin-1(Fer-1)是一種有效的、選擇性的鐵死亡抑制劑。HT-1080細(xì)胞中,Fer-1(0.5μM)抑制鐵死亡激活劑Erastin(Cat#:NBS5879,EC50=60nM)和RSL3誘導(dǎo)的細(xì)胞死亡,但對過氧化氫、魚藤酮,或星孢菌素誘導(dǎo)的細(xì)胞死亡沒有效果。Fer-1抑制Erastin誘導(dǎo)的活性氧生成和脂質(zhì)過氧化。在基于細(xì)胞水平的亨廷頓病、腦室周圍白質(zhì)軟化癥(PVL)和近端腎小管損傷模型中,Fer-1以劑量依賴性的方式提高細(xì)胞存活率。大鼠海馬器官型腦片培養(yǎng)模型中,Fer-1(2μM)抑制L-谷氨酸鹽誘導(dǎo)的細(xì)胞死亡。Fer-1還能通過捕獲過氧自由基的方式阻抑脂質(zhì)自氧化。

實(shí)驗(yàn)方法(Assay):HT-22 cells were seeded in 6-well culture plate (2 × 105 cells/well). Glutamate (1.25, 2.5, 5, 10, 20 mM) was applied to HT-22 cells for 12, 24, 36, and 48 hours. HT-22 cells were cultured with different concentrations of ferrostatin-1 (3, 6 and 12μM) for 16 hours prior to exposure to 5 mM glutamate. After 24 hours, the subsequent analyses were conducted.
結(jié)果說明(Result):Ferrostatin-1 dose-dependently reduced cell death, with a maximum effect at a dose of 12 μM, but had no significant effect on the survival of HT-22 cells deprived of glutamate.

動物模型(Animal Model):Male C57BL/6 mice
實(shí)驗(yàn)方法(Assay):The LPS-induced ALI model was induced by instilling intratracheally 50 μl of LPS solution (0.2 g/L), then Fer-1 (0.8 mg/kg) was administered after LPS challenge via tail vein injection. The Fer-1 was dissolved in DMSO first, and diluted with 0.9% NaCl. The final concentration of Fer-1 and DMSO was 0.2 mg/ml and 0.1% respectively. After the treatments for 16 h, the mice in each group were euthanized and bronchoalveolar lavage (BAL) fluid was collected via lung lavage.
