硫化氫(H2S)熒光探針產(chǎn)品專題—WSP-1/WSP-5/AzMC
背景描述
硫化氫(H2S)是第三大氣體介質(zhì),另兩種是一氧化氮(NO)和一氧化碳(CO),在哺乳動物的免疫學、神經(jīng)學、心血管和呼吸系統(tǒng)中發(fā)揮顯著的生理效應(yīng)。哺乳動物體內(nèi)H2S歸因于至少三種內(nèi)源性酶:胱硫醚-β-合成酶(Cystathionine-β-synthase,CBS),細胞胱硫醚-γ-裂解酶(cystathionine γ-lyase,CGL,CSE),3-巰基丙酮酸硫基轉(zhuǎn)移酶(3-mercaptopyruvate sulfurtransferase,3-MST),這些酶在不同的器官和組織內(nèi),使用半胱氨酸或半胱氨酸衍生物為底物,并將其轉(zhuǎn)化為H2S。
H2S作為一種高度反應(yīng)性分子,能夠與大量的生物靶標反應(yīng),這些反應(yīng)涉及到H2S的各種生理功能。比如,H2S立即與高鐵血紅蛋白反應(yīng)生成硫化血紅蛋白,后者可用作H2S的代謝庫。H2S是一種強效的還原劑,很可能與內(nèi)源性氧化劑如過氧亞硝基陰離子,超氧化物和過氧化氫反應(yīng)。能引起蛋白硫巰基化修飾(-SSH),這是一種重要的蛋白翻譯后修飾,提供一種可能機制使得H2S改變細胞內(nèi)大量的蛋白和酶的功能。盡管如此,內(nèi)源性H2S的生成以及外源注射H2S對許多疾病都表現(xiàn)出保護效應(yīng),為此,了解H2S的化學和特征,以及開發(fā)一種有效且方便的方法來檢測生物體系內(nèi)H2S顯得十分重要。

產(chǎn)品特點
我公司(上海諾寧生物)提供一系列新開發(fā)的高效且便捷的硫化氫(H2S)熒光探針——Washington State Probe(WSP)(WSP-1, WSP-2, WSP-3, WSP-4, WSP-5),這類探針由M. Xian團隊開發(fā)并經(jīng)多方驗證非常適合生物樣本的H2S檢測。這類探針的特點如下:
1)低背景:探針本身熒光背景非常低,這對H2S的高靈敏檢測至關(guān)重要。這些探針的羥基基團被酯化,因此呈現(xiàn)出弱熒光和弱的量子產(chǎn)量(Φf < 0.1,見下表1)。

2)靈敏度:探針能與H2S快速結(jié)合熒光信號顯著加強,僅幾分鐘熒光信號急劇增加且達到穩(wěn)態(tài)(見表2)。在梯度濃度的H2S體系中,最大發(fā)射波長下的熒光信號與濃度呈現(xiàn)良好的線性關(guān)系。

3)選擇性:探針表現(xiàn)出良好的H2S特性行結(jié)合,與包括Cys和GSH在內(nèi)的反應(yīng)性硫類物質(zhì)相比(見Fig 1)。

4)穩(wěn)定性:在細胞內(nèi)酯酶(esterase E-0887,from rabbit liver)存在的情況下探針表現(xiàn)
出良好的穩(wěn)定性。適合活細胞熒光成像研究(見Fig 2)。

我公司(上海諾寧生物)提供一種基于香豆素的硫化氫(H2S)熒光探針—7-Azido-4
-methylcoumarin (AzMC、C-7Az),這款探針由XJ Tang團隊開發(fā)并經(jīng)多方驗證非常適合生物樣本的H2S檢測。此探針特點如下:

1) 與硫化氫反應(yīng)的工作原理

2) 低背景和高靈敏度
C-7Az在脫氣的PBS緩沖液(10mM,pH 7.4)中無熒光,一旦加入NaHS(H2S供體),C-7Az即可開啟熒光反應(yīng),最大吸收峰呈現(xiàn)~15nm的紅光遷移,在340nm下激發(fā)發(fā)生顯著的熒光增強。如Fig 5所示,加入30μM NaHS(H2S供體)10min后,熒光增加高達36倍;加入30μM NaHS(H2S供體)60min后熒光增強108倍。

3) 高選擇性
與其它陰離子和生物線管的反應(yīng)硫化物(RSS)、反應(yīng)氧物質(zhì)(ROS)和反應(yīng)氮物質(zhì)(RNS)相比,C-7Az高選擇性結(jié)合硫化氫。如Fig 6所示,加入100μM C-7Az,含100μM NaHS的體系內(nèi)445nm處的相對熒光強度,比其它1mM相關(guān)的RSS、ROS、RNS和陰離子體系高31-60倍。

4) 良好的線性關(guān)系
C-7Az在脫氣的PBS緩沖液和各種濃度的胎牛血清(0.25-100μM)中與H2S反應(yīng),呈現(xiàn)出良好的線性關(guān)系(如Fig 7)。

5) 適用于細胞內(nèi)H2S檢測(Fig 8)。
